Receiving and inspecting a vial

Inspect the seal and cap for damage on arrival, confirm the lot number matches your paperwork, and record the date received. A lyophilised cake may appear as a compact disc, a thin film or loose powder — all are normal. Transfer to -20°C storage promptly and avoid leaving vials at ambient temperature longer than necessary.

Storage of lyophilised material

Sealed lyophilised vials are most stable at -20°C, shielded from light and moisture. Keep vials upright in their original packaging. Repeated temperature cycling is the most common cause of degradation in a research setting, so decant working stock rather than returning the same vial to room temperature repeatedly.

Reconstitution technique

Add your chosen sterile diluent slowly against the inner wall of the vial so the stream does not strike the cake directly. Allow the material to dissolve without shaking; gentle swirling is sufficient. Vigorous agitation introduces shear stress and foaming, both of which can damage peptide structure. Solutions should be clear and free of visible particulates.

Working solutions and stability

Once in solution, refrigerate at 2-8°C and protect from light. Aliquot into single-use volumes to avoid freeze-thaw cycling. Label every aliquot with compound, concentration, diluent, date prepared and lot number so results remain traceable back to the analytical certificate.

Documentation and traceability

Good research records tie every observation back to a lot number. Record the certificate of analysis reference, purity figure, preparation date and storage history alongside your experimental data. This is what makes a result reproducible and defensible.

Handle all materials with appropriate personal protective equipment in a controlled laboratory environment. Treat unused solutions and empty vials as laboratory waste and dispose of them according to your institution's protocols and local Australian regulations.
